Establishment of Clonal Cell Lines from Human Lung Cancer Cell Line which Produces Human Chorionic Gonadotropin in Vitro
Kim Choo-Soung
Abstract
The ChaGo cell line, derived froma human lung carcinoma produces human chorionic gonadotropin in vitro. A clonal line has been derived from the ChaGo cultures by isolating a single cell in a capillary tube and observing its growth until adequate numbers of cells were present for snbculture. The clonal line has growth patterns similar to that of the uncloned cultures and both have popula
tion doubling times of 24-30 hours. Cytogenetic studies show that have a predominance of cells with 51 to 54 chromosomes but the range is more restricted in the clone. Studies of production of the free alpha subunit of human chorionic gonadotropin (hCG) have been completed and while after 7 days the uncloned cultures produced 500 nanograms/ml, comparable cultures of the clone produced 1000 nanograms/ml. Studies of production of beta subunit and complete hCG are in progress.
